History

Designed by Baar and colleagues at Erasmus University Medical Center from the FOXO4 domain that binds p53, synthesized with D-amino acids in reverse order so the peptide resists proteolysis while retaining binding activity.

What retro-inverso means, and why it is the point

FOXO4-DRI is not a conventional peptide analogue. The name records its construction: D-amino acids, Retro-Inverso. The sequence is reversed and every residue is built from the D-enantiomer rather than the L-form found in natural proteins.

Running both changes together is what makes the design work. Reversing the sequence while inverting the chirality leaves the side-chain topology approximately where it was, so the molecule still presents the interaction surface it needs to — but proteases, which recognise L-amino-acid backbones, largely do not act on it. The result is a peptide that keeps the binding geometry and loses most of the degradation.

Functionally it competes with endogenous FOXO4 for p53 binding. In senescent cells FOXO4 sequesters p53 in PML nuclear bodies; displacing it is the mechanism behind the compound's use as a senolytic research tool.

This matters for ordering because a conventionally synthesised L-form of the same sequence is a different reagent that will not behave the same way. Identity confirmation on the Certificate of Analysis should be read with that in mind.
